RMZ-RC2
- Main features:
RMZ is a human alveolar rhabdomyosarcoma cell line originally
established in our laboratory from a bone marrow metastasis. RMZ
is able to differentiate along the myogenic pathway in
vitro; most cells express desmin, a minority express
embryonic myosin (right picture above) and give rise to
multinuclear myotube-like elements. Clone RC2 maintains the
differentiative properties of the RMZ cell line while growing
faster.
- Culture conditions:
- Medium: Dulbecco's MEM + 10% FCS.
- Split when confluent, usually once every 4-5 days. Split
ratio 1:2 - 1:3; don't dilute beyond 1:3 or cells will
stop growing. Detach with Trypsin-EDTA for 2' - 3' at room
temperature, then stop trypsin with DMEM+FCS and centrifuge.
- Freezing medium: 90% FCS (or NCS), 10% DMSO; cell density for
freezing: 2 vials from a confluent 25 cm² flask.
- Thawing: seed one 25 cm² flasks from one vial.
- Warning: freezing of these cells is somewhat difficult. To be
on the safe side do not freeze all your cultures at once.
Maintain some flasks in continuous culture until you check that
thawing yields viable and proliferating cells.
