CCA
- Main features:
CCA is a human embryonal rhabdomyosarcoma cell line established
in our laboratory. CCA cells express desmin, a minority of cells
also express embryonic myosin and display an elongated shape,
however somatic fusion was not observed in vitro.
- Culture conditions:
- Medium: Dulbecco's MEM + 10% FCS.
- Split only when extremely confluent. We use the
following rule of thumb: CCA is ready only if one sees with the
naked eye thick vortexes of cells (we call this "brushwork").
Split ratio 1:2 - 1:6. Detach with trypsin-EDTA for 2' - 3' at
37°C, then stop trypsin with DMEM+FCS and centrifuge.
- Freezing medium: 90% FCS (or NCS), 10% DMSO; cell density for
freezing: 3 vials from an overconfluent 25 cm² flask.
- Thawing: seed two 25 cm² flasks from one vial.
