B78H1
- Main features:
B78H1 is an amelanotic clone derived from the murine melanoma B16
in the laboratory of Selma Silagi. Usually B16 cells have a low
expression of class I histocompatibility antigens (shaded FACS
profile above), and show a rapid induction after interferon-gamma
treatment (solid profile). Unlike B16, B78H1 cells are class I
negative even after interferon-gamma induction.
- Culture conditions:
- Medium: Dulbecco's MEM + 10% FCS.
- Split ratio 1:5 - 1:10. Detach with Trypsin-EDTA for 5' - 6'
at 37°, then stop trypsin with DMEM+FCS and centrifuge.
- Freezing medium: 90% FCS (or NCS), 10% DMSO; cell density for
freezing: 3-4 vials from a confluent 25 cm² flask.
- Thawing: seed one 25 cm² flask from one vial.
